How does your risk of ruin change as your bankroll fluctuates? If you start with a $3000 roll and win $2000, do you now have the same ROR of someone that started with a $5000 BR? I've heard many times that bet strategy should be recalculated after winning or losing significantly but shouldn't win rate balance out regardless?

your risk of ruin drops. You can maintain the same level of risk as your original bankroll by increasing your unit size. Many players get stung by this (they go on a losing streak with the new bigger bets) so be very careful and considerate when making the decision to up your unit size.
As your bankroll gets bigger it becomes harder and harder to replace and therefore players with huge bankrolls usually choose to play to an extremely low level of risk.
